Prime Editing searches to find the precise place in the genome to edit and replaces the segment of faulty DNA with a correct copy of DNA.  A single Prime Editor can correct the individual mutations found across patients, meaning Prime Editing can potentially address more than 90 percent of known disease-causing genetic mutations.

Position Overview

We are currently seeking a highly talented and motivated Scientist/Senior Associate Scientist with great team spirit to join the New Opportunities Team at Prime Medicine. In this position, the individual will support the development of Prime Editing therapies to treat rare genetic lung diseases with high unmet need. The main responsibilities will include, but are not limited to, molecular cloning, and delivering Prime Editors into human assessing gene editing frequencies using NGS and characterizing the phenotypes of the edited cells. A successful candidate will have strong knowledge and technical skills in molecular biology and mammalian cell biology. Additional experience in gene editing, screening, next generation sequencing prep/analysis and the pathobiology of disease is a plus. The ideal candidate is hard-working, highly collaborative and organized, with great enthusiasm for science.

Primary Responsibilities

  • Perform various molecular biology techniques (e.g. molecular cloning. Gibson Assembly, Golden Gate Assembly, transformation, plasmid prep, primer design, PCR, agarose gel electrophoresis, DNA/RNA purification, etc.) to prepare research materials.
  • Maintain mammalian cell culture (e.g. cell lines, iPSCs) and deliver Prime Editors to the cells with different methods (e.g. transfection,  LNP-mediated transduction).
  • Extract DNA/RNA/protein from cells for downstream analyses including, but not limited to, next-generation sequencing, qPCR and western blot.
  • Keep meticulous experimental records, documented in electronic lab notebooks.
  • Analyze, summarize and present experimental data,
  • Adhere to all laboratory safety requirements and procedures, assist in maintaining general laboratory functionality.

Qualifications

  • A PhD/MS in Biology/Engineering or related discipline with ideally 3+ years of experience with molecular biology, biochemistry and cell biology.
    • Biotech / Pharma experience is a plus
  • Proficiency in molecular biology as described above
  • Proficiency in mammalian cell culture and transfection.
  • Working knowledge of gene editing tools and principles (ZFN, TALE, CRISPR or DNA base editors) is a plus.
  • Experience developing cell-based assays including assays of transporter or channel function, and other cell function assays
  • Experience in lung biology and disease models is a plus.
  • Proficiency with DNA analysis software (e.g. Snapgene, Geneious, Primer 3), data analysis software (e.g. Graphpad Prism), Microsoft Office products (Word, Excel, PowerPoint) and literature searching tools (e.g. PubMed).
  • Demonstrated maturity and scientific contributions within a complex matrixed organization.
  • Outstanding interpersonal and communication skills, self-awareness, and ability to work in a highly collaborative, fast-paced environment.
